Biological Background: ST6GAL1 Function and Localization
- ST6GAL1 encodes beta-galactoside alpha-2,6-sialyltransferase 1 (ST6Gal I, SIAT1, CDw75), a type II membrane protein resident in the Golgi apparatus.
- Catalyzes the transfer of sialic acid from CMP-sialic acid to terminal galactose residues on N-glycans, producing α2,6-linked sialylated glycoconjugates.
- In B lymphocytes, ST6GAL1 generates the CDw75, HB-6 and CD76 cell-surface differentiation antigens.

PMID:1730763 - Mediates sialylation of the Fc region of IgG antibodies, preferentially on the α1,3 mannose antenna, conferring anti-inflammatory properties.
- Located in the Golgi stack membrane, with a cleaved soluble form also secreted.
- Undergoes alternative splicing, producing multiple isoforms; annotated as a glycoprotein and phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal luminal domain (aa 27-200), which may detect multiple ST6GAL1 isoforms. In Western blot, confirm the band size (~47 kDa) under reducing conditions.
- For IHC-P, antigen retrieval protocols (e.g., heat-induced epitope retrieval) and appropriate tissue controls are recommended to optimize staining specificity.
- In ELISA, use recombinant ST6GAL1 protein or peptide as coating antigen; cross-reactivity with mouse and rat orthologs enables comparative studies.
- Avoid using samples with high endogenous IgG if detecting IgG-sialylated targets, as this may cause interference.
