Biological Background: ST3GAL5 Function and Localization
- The ST3GAL5 gene encodes the enzyme lactosylceramide alpha-2,3-sialyltransferase, commonly known as GM3 synthase, ST3Gal V, or sialyltransferase 9 (SIAT9).
- It catalyzes the transfer of N-acetylneuraminic acid (NeuAc) from CMP-NeuAc to the terminal galactose of lactosylceramide (LacCer) to form ganglioside GM3, a critical step in glycosphingolipid biosynthesis that regulates multiple cellular processes including proliferation, differentiation, and apoptosis. - The enzyme preferentially uses LacCer but can also sialylate other glycolipids such as galactosylceramide (GalCer), asialo-GM2 (GA2), and asialo-GM1 (GA1) with lower efficiency.
- ST3GAL5 mRNA is ubiquitously expressed, with highest levels in the brain (cerebral cortex, temporal lobe, putamen), skeletal muscle, placenta, and testis.
- Mutations in ST3GAL5 are linked to severe neurological disorders, including infantile-onset epileptic encephalopathy and intellectual disability, underscoring its essential role in brain development.
- The protein is localized to the Golgi apparatus membrane, contains a signal-anchor transmembrane domain, forms disulfide bonds, and undergoes alternative splicing to generate multiple isoforms.
Experimental Guidance and Technical Tips
- For Western blot, use denaturing conditions; the predicted molecular weight is approximately 48 kDa, but monitor for possible dimerization or post-translational modifications.
- Validate the antibody specificity with appropriate positive and negative controls (e.g., cell lines with known expression, siRNA knockdown, or knockout lysates) in your experimental system.
- For ELISA, titrate the antibody to determine optimal working concentration and assess cross-reactivity with related sialyltransferases if necessary.
