Biological Background: ST3GAL4 Function and Localization
- ST3GAL4 encodes a beta-galactoside alpha-2,3-sialyltransferase that catalyzes the transfer of sialic acid from CMP-Neu5Ac to Galbeta-(1->3)-GalNAc and Galbeta-(1->4)-GlcNAc-terminated glycoconjugates, mediating terminal sialylation of glycoproteins and glycolipids.

PMID:8611500 - Plays a key role in hemostasis by sialylating plasma von Willebrand factor, thereby preventing its recognition by asialoglycoprotein receptors and subsequent clearance, and also regulates ASGPR-mediated platelet clearance.
- Participates in the biosynthesis of sialyl Lewis X epitopes on both O- and N-glycans, which serve as ligands for selectins, and is essential for selectin-mediated leukocyte rolling and adhesion during extravasation.

PMID:25498912 - In glycosphingolipid metabolism, sialylates GM1 and GA1 gangliosides to form GD1a and GM1b, respectively, and synthesizes ganglioside LM1, a major structural component of peripheral nerve myelin.

PMID:8611500 - Localizes primarily to the Golgi stack membrane and can also be secreted.
- Highly expressed in adult placenta, heart, and kidney.
- Features alternative splicing, disulfide bonds, and glycosylation; belongs to the glycosyltransferase 29 family.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 234-333 of human ST3GAL4 (isoform NP_001241686.1). This region may be conserved across isoforms; consider sequence alignment to verify specificity.
- Validated for Western blot (WB) and ELISA applications. For WB, a band around 36-38 kDa is expected; adjust electrophoresis conditions to resolve potential isoforms.
- Cross-reactivity with mouse has been confirmed; validate in the relevant sample system for your experimental design.
- As a polyclonal antibody, batch-to-batch variability may occur; include appropriate positive and negative controls.
