Biological Background: ST3GAL4 Function and Localization

  • ST3GAL4 encodes CMP-N-acetylneuraminate-beta-galactosamide-alpha-2,3-sialyltransferase 4 (alpha 2,3-sialyltransferase IV), also known as SAT-3, ST-4, ST3GalIV, and STZ, a key member of the sialyltransferase family.
  • It catalyzes the transfer of sialic acid (Neu5Ac) from CMP-Neu5Ac onto Galbeta-(1->3)-GalNAc- and Galbeta-(1->4)-GlcNAc-terminated glycoconjugates through an alpha2-3 linkage, mediating terminal sialylation of glycoproteins and glycolipids (PubMed:8288606, PubMed:8611500, PubMed:37632720).   • Plays a major role in hemostasis by sialylating plasma von Willebrand factor (VWF), which prevents its recognition by asialoglycoprotein receptors (ASGPR) and subsequent clearance; also regulates ASGPR-mediated platelet clearance.
  • Essential for selectin-mediated leukocyte rolling and adhesion: participates in the biosynthesis of sialyl Lewis X epitopes on O- and N-glycans, recognized by SELE, SELP, and SELL during extravasation (PubMed:25498912). Related references: PMID:25498912
  • In glycosphingolipid biosynthesis, sialylates GM1 and GA1 gangliosides to form GD1a and GM1b, respectively (PubMed:8288606); also metabolizes brain c-series ganglioside GT1c to GQ1c and synthesizes ganglioside LM1, a major structural component of peripheral nerve myelin (PubMed:8611500).
