Biological Background: ST3GAL1 Function and Localization

  • ST3GAL1 (CMP-N-acetylneuraminate-beta-galactosamide-alpha-2,3-sialyltransferase 1), also known as SIAT4A or ST3Gal I, is a member of the glycosyltransferase family involved in glycoprotein and glycolipid sialylation.

  • ST3GAL1 specifically adds sialic acid to core 1 O-glycans (Galbeta-(1->3)-GalNAc-O-Ser/Thr), the major structure of mucin-type O-glycans, thereby modifying glycoproteins such as SPN/CD43 and PTPRC/CD45 on T cells.
  • Through sialylation of T cell glycoproteins, ST3GAL1 plays a homeostatic role in regulating CD8-positive T cell numbers by preventing premature apoptosis in thymic T cells and controlling survival of memory T cells in secondary lymphoid organs.

  • The enzyme localizes to the Golgi apparatus, specifically the Golgi stack membrane and trans-Golgi network membrane; a soluble secreted form has also been identified.
  • Expression of ST3GAL1 is widespread, with highest levels detected in lung, liver, skeletal muscle, kidney, pancreas, spleen, and placenta.
