Biological Background: ST3GAL1 Function and Localization
- ST3GAL1 (CMP-N-acetylneuraminate-beta-galactosamide-alpha-2,3-sialyltransferase 1), also known as SIAT4A or ST3Gal I, is a member of the glycosyltransferase family involved in glycoprotein and glycolipid sialylation.
- It catalyzes the transfer of sialic acid (Neu5Ac) from CMP-Neu5Ac to Galbeta-(1->3)-GalNAc-terminated glycoconjugates via an alpha2-3 linkage, a key step in terminal sialylation. Related references: PMID:31784620 PMID:8027041 PMID:31719620
- ST3GAL1 specifically adds sialic acid to core 1 O-glycans (Galbeta-(1->3)-GalNAc-O-Ser/Thr), the major structure of mucin-type O-glycans, thereby modifying glycoproteins such as SPN/CD43 and PTPRC/CD45 on T cells.
- Through sialylation of T cell glycoproteins, ST3GAL1 plays a homeostatic role in regulating CD8-positive T cell numbers by preventing premature apoptosis in thymic T cells and controlling survival of memory T cells in secondary lymphoid organs.
- It also acts on gangliosides, converting GM1a to GD1a, GA1 to GM1b, and GD1b to GT1b. Related references: PMID:8027041
- The enzyme localizes to the Golgi apparatus, specifically the Golgi stack membrane and trans-Golgi network membrane; a soluble secreted form has also been identified.
- Expression of ST3GAL1 is widespread, with highest levels detected in lung, liver, skeletal muscle, kidney, pancreas, spleen, and placenta.
- The protein is a transmembrane glycoprotein with a signal-anchor, containing disulfide bonds and a short cytoplasmic tail.

- For Western blot (WB), consider using a dilution range of 1:500 to 1:2000 and validate with positive control lysates from tissues known to express ST3GAL1, such as lung or liver.

- As the antibody shows cross-reactivity with mouse and rat, it can be employed for orthologous studies, but always confirm specificity in the relevant species context.
