  • SSRP1 (FACT80, T160) is a subunit of the Facilitates Chromatin Transcription (FACT) complex, a general chromatin factor that reorganizes nucleosomes to facilitate DNA-templated processes including transcription elongation, DNA replication, and DNA repair.
  • The FACT complex functions as a histone chaperone: during transcription, it promotes the transient dissociation of one H2A-H2B dimer from the nucleosome to allow RNA polymerase II passage, then reassembles the nucleosome afterward.
  • SSRP1 binds specifically to double-stranded DNA and shows low-affinity binding to cisplatin-modified DNA; this interaction may potentiate cisplatin-induced cell death by blocking replication and repair of damaged DNA.
  • It acts as a transcriptional coactivator for p63/TP63 and may contribute to p53/TP53 phosphorylation at Ser-392 via an association with casein kinase II (CK2).
