Biological Background: SSNA1 Function and Localization
- SSNA1 (also known as NA14) is a microtubule nucleation factor that directly binds to and stabilizes dynamic microtubules, slowing their growth and shrinkage at both plus and minus ends, and protects them from severing by the SPAST enzyme. Related references: PMID:34970964
- It induces microtubule branching by forming long SSNA1 fibrils that guide protofilaments to split apart from the mother microtubule and form daughter microtubules.
- SSNA1 is required for proper axon outgrowth and branching during neurogenesis. Related references: PMID:25390646
- It plays an essential role in cell division. Related references: PMID:25390646
- The protein localizes to multiple cellular structures including the nucleus, cytoplasm, centrosome, centrioles, midbody, and the basal body/axoneme of cilia and flagella.
- SSNA1 is widely expressed across tissues and undergoes post-translational acetylation.
- Originally identified as a nuclear autoantigen in Sjögren's syndrome, SSNA1 contains a coiled-coil domain.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to the N-terminal region (aa 1-119) of human SSNA1; when developing new detection reagents, consider using the full-length protein or conserved regions to maintain cross-reactivity with mouse and rat.
- For Western blotting (WB), the predicted molecular weight is approximately 14 kDa, but post-translational modifications may shift the observed band; validate in the relevant sample system and consider using positive controls.
- In immunohistochemistry with paraffin-embedded sections (IHC-P), SSNA1's varying subcellular localization (nucleus, centrosome, midbody) may require optimization of antigen retrieval and blocking conditions.
- When performing ELISA, titrate the antibody to determine the optimal coating and detection conditions for your specific assay format.
