Biological Background: SRSF3 Function and Localization
- SRSF3 (also known as SFRS3 or SRp20) is a member of the serine/arginine-rich (SR) protein family of pre-mRNA splicing factors.
- It specifically binds the consensus motif 5'-C[ACU][AU]C[ACU][AC]C-3' within pre-mRNA to promote inclusion of target exons during alternative splicing. Related references: PMID:17036044, PMID:26876937, PMID:32440474
- Interaction with YTHDC1, an m6A reader, recruits SRSF3 to mRNA regions adjacent to m6A sites, coupling m6A modification to splicing regulation. Related references: PMID:26876937
- SRSF3 also functions as a nuclear export adapter; it binds mature mRNA and facilitates transfer to the NXF1-NXT1 (TAP/NXF1) export receptor to enhance mRNA nuclear export. Related references: PMID:11336712, PMID:18364396
- Through YTHDC1, SRSF3 participates in the selective nuclear export of m6A-modified mRNAs. Related references: PMID:28984244
- It localizes predominantly to the nucleus and nuclear speckles, but can also be found in the cytoplasm in the context of mRNA transport.
- SRSF3 undergoes several post-translational modifications, including phosphorylation and acetylation, which regulate its activity and interactions.
- The protein contains RNA recognition motifs (RRMs) and a characteristic arginine/serine-rich (RS) domain common to SR proteins.
Experimental Guidance and Technical Tips
- The immunogen used to generate this recombinant monoclonal antibody is a synthetic peptide spanning amino acids 1–100 of human SRSF3. This N‑terminal region is conserved and may allow the antibody to recognize multiple isoforms; however, validation in the target sample context is recommended.
- For western blotting (WB), the anticipated band for SRSF3 is approximately 19 kDa. Include appropriate positive and negative controls, and consider testing different lysate preparation and loading conditions to optimize signal.
- In immunohistochemistry (IHC‑P), SRSF3 staining is expected primarily in the nucleus, with possible cytoplasmic signal due to its role in mRNA export. Validate the antibody in tissues known to express SRSF3 and include isotype controls.
- For ELISA applications, titrate the antibody and use a relevant antigen standard to determine sensitivity and specificity; conditions should be established empirically for each assay format.
