Biological Background: SRSF2 Function and Localization
- SRSF2, also known as SC35 or PR264, is a key serine/arginine-rich (SR) splicing factor essential for pre-mRNA processing.
- It is necessary for the formation of the earliest ATP-dependent splicing complex and bridges U1 snRNP (5’ splice site) and U2AF (3’ splice site) during spliceosome assembly.
- The protein binds to purine-rich RNA sequences, such as 5’-AGSAGAGTA-3’ and 5’-GTTCGAGTA-3’, and commits specific transcripts like beta-globin mRNA to the splicing pathway.
- SRSF2 is localized in the nucleus, predominantly in nuclear speckles and the nucleoplasm, consistent with its role in mRNA processing.
- Phosphorylation by SRPK2 regulates its splicing activity and is required for cisplatin-induced apoptosis; acetylation is also associated with its regulation.
- As a member of the SR protein family, SRSF2 is subject to alternative splicing itself, producing multiple isoforms (e.g., SFRS2A, SRp30b).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide within the N-terminal region (aa 1-100), which may detect various SRSF2 isoforms; confirm specificity by siRNA knockdown or using isoform-specific controls.
- For Western blotting, a band around 25 kDa is expected; validate in the relevant sample system and under reducing conditions.
- For IHC-P, consider antigen retrieval methods and validate in formalin-fixed paraffin-embedded tissue sections from human, mouse, or rat.
- Cross-reactivity with mouse and rat has been reported; sequence alignment of the immunogen region across species may support this, but independent validation is recommended.
