- SRSF1, also known as ASF/SF2, is a prototypical serine/arginine-rich (SR) protein that belongs to the SR protein family of splicing factors.
- It plays a critical role in pre-mRNA splicing by preventing exon skipping and ensuring splicing accuracy; it bridges U1 snRNP and U2AF via its RS domain.
- SRSF1 binds purine-rich RNA sequences, such as the octamer 5'-RGAAGAAC-3' or decamers AGGACAGAGC/AGGACGAAGC, and three octamer copies form a powerful splicing enhancer (ASE).
- Alternative splicing generates isoforms ASF-2 and ASF-3, which function as splicing repressors rather than activators.
- In addition to splicing regulation, SRSF1 may act as an mRNA export adapter, facilitating nuclear export through the TAP/NXF1 pathway.
- Subcellular localization includes nuclear speckles (primary), cytoplasm, and chromatin, consistent with its dynamic roles in RNA processing and transport.
- The protein undergoes multiple post-translational modifications, including phosphorylation, methylation, and acetylation, which modulate its activity and interactions.

- The immunogen spans the N-terminal region (a.a. 1-248) of human SRSF1, so the antibody may recognize all known isoforms; however, isoform-specific reactivity should be validated.
- For Western blot, the predicted molecular weight is approximately 28 kDa, but phosphorylated forms may migrate slightly slower; consider using phosphatase inhibitors in lysis buffers.
- For ELISA, the antibody may be used to detect purified recombinant SRSF1 or endogenous protein in cell lysates; optimization of coating and detection conditions is recommended.
- Cross-reactivity with mouse and rat orthologs is indicated; confirm performance in your specific sample system before large-scale use.
