Biological Background: SRPK1 Function and Localization
- SRPK1 is an ATP-dependent serine/threonine protein kinase that specifically phosphorylates SR splicing factors at serine residues within RS domains, regulating pre-mRNA splicing.
- It controls the intranuclear distribution of splicing factors in interphase cells and the reorganization of nuclear speckles during mitosis.
- Beyond splicing, SRPK1 influences mRNA maturation, chromatin reorganization in somatic and sperm cells, and cell cycle progression.
- Isoform 2 phosphorylates substrates including SFRS2, ZRSR2, LBR, and PRM1, mediates HBV core protein phosphorylation, and negatively regulates HBV replication by reducing pgRNA packaging efficiency.
- Isoform 1 and isoform 2 both promote splicing of exon 10 in MAPT/TAU, and their ratio dictates cell fate in K-562 leukemic cells (isoform 2 supports proliferation, isoform 1 differentiation).
