Biological Background: SRPK1 Function and Localization

  • SRPK1 is an ATP-dependent serine/threonine protein kinase that specifically phosphorylates SR splicing factors at serine residues within RS domains, regulating pre-mRNA splicing.
  • It controls the intranuclear distribution of splicing factors in interphase cells and the reorganization of nuclear speckles during mitosis.
  • Beyond splicing, SRPK1 influences mRNA maturation, chromatin reorganization in somatic and sperm cells, and cell cycle progression.
  • Isoform 2 phosphorylates substrates including SFRS2, ZRSR2, LBR, and PRM1, mediates HBV core protein phosphorylation, and negatively regulates HBV replication by reducing pgRNA packaging efficiency.
  • Isoform 1 and isoform 2 both promote splicing of exon 10 in MAPT/TAU, and their ratio dictates cell fate in K-562 leukemic cells (isoform 2 supports proliferation, isoform 1 differentiation).
  • SRPK1 phosphorylates the N-terminus of ERC1. Related references: PMID:30745339
  • The protein is localized to the cytoplasm, nucleus, nucleoplasm, nuclear speckles, chromosomes, and microsomes.
  • Tissue-specific expression: isoform 2 is predominant in testis and also present at lower levels in several other tissues; isoform 1 is testis-specific and less abundant.
  • SRPK1 is a phosphoprotein that belongs to the protein kinase superfamily and contains an ATP-binding pocket.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 290–480 of human SRPK1 (NP_003128.3). Consider verifying the sequence conservation if using in mouse or rat models.
  • For Western blot, the target protein weight is approximately 74 kDa. Include appropriate loading controls and optimize blocking conditions.
  • Immunofluorescence/ICC experiments should account for SRPK1’s nuclear and cytoplasmic localization; standard fixation and permeabilization may be suitable.
  • ELISA protocols may benefit from peptide-coated plates or a sandwich format; pair with a matched detection antibody if needed.
  • Validate antibody performance in the relevant sample system, as cross-reactivity has been demonstrated with human, mouse, and rat.
