Biological Background: SRPK1 Function and Localization
- SRPK1 (SFRS protein kinase 1) is a serine/arginine-rich protein-specific kinase from the serine/threonine kinase family.
- It specifically phosphorylates SR splicing factors on serine residues within RS domains, regulating pre-mRNA splicing and intranuclear distribution of splicing factors.
- Beyond splicing, SRPK1 influences mRNA maturation, chromatin reorganization in somatic and sperm cells, and cell cycle progression.
- Isoform 2 employs a directional, dual-track phosphorylation mechanism on SRSF1, involving processive and distributive steps mediated by a docking groove.
- SRPK1 is present in multiple subcellular locations: cytoplasm, nucleus, nucleoplasm, nuclear speckles, and chromosomes.
- Two isoforms exist: isoform 1 (mainly testis-specific) and isoform 2 (testis and broadly in tissues like heart, ovary, small intestine, liver, kidney, pancreas, skeletal muscle).
- SRPK1 isoform 2 mediates HBV core protein phosphorylation and negatively regulates HBV replication by reducing pgRNA packaging, independent of core phosphorylation.
