Biological Background: SRP14 Function and Localization
- SRP14, also known as the 18 kDa Alu RNA-binding protein, is a core component of the signal recognition particle (SRP), a universally conserved ribonucleoprotein complex that mediates the cotranslational targeting of secretory and membrane proteins to the endoplasmic reticulum (ER). - Together with SRP9 and the Alu domain of SRP RNA, SRP14 forms the elongation arrest domain of SRP, which transiently pauses translation to ensure efficient protein translocation.
- The SRP9/SRP14 heterodimer is essential for high-affinity binding to SRP RNA, a critical step for assembly of a functional SRP particle.
- SRP14 is primarily localized in the cytoplasm, consistent with its role in targeting nascent polypeptide chains to the ER membrane.
- The protein undergoes phosphorylation (as indicated by mass spectrometry–based proteomics), suggesting potential regulation by post-translational modification.
- The theoretical molecular weight of SRP14 is approximately 14.6 kDa, consistent with the observed band at 15 kDa in Western blotting experiments.

- The immunogen used to generate this antibody was a recombinant fusion protein covering near full-length human SRP14 (amino acids 1–136). This design favors recognition of native, folded epitopes.
- For Western blot detection, consider using whole-cell lysates from human, mouse, or rat cell lines, as SRP14 is ubiquitously expressed. A band near 15 kDa is expected.
