  • Serglycin (also known as hematopoietic proteoglycan core protein, platelet proteoglycan core protein, or P.PG) is encoded by the SRGN gene and belongs to the proteoglycan family.
  • It plays a critical role in the formation of mast cell secretory granules and mediates the storage of various compounds within secretory vesicles.
  • Serglycin is essential for the storage of certain proteases in both connective tissue and mucosal mast cells, as well as for granzyme B storage in T-lymphocytes.
  • In neutrophils, it contributes to the localization of neutrophil elastase within azurophil granules.
  • The protein mediates the processing of matrix metalloproteinase 2 (MMP2) and regulates the secretion of tumor necrosis factor (TNF) and other proteases.
  • Serglycin participates in cytotoxic cell granule-mediated apoptosis by forming a complex with granzyme B that is delivered to target cells via perforin.

  • The immunogen corresponds to amino acids 28–158 of human SRGN, excluding the N-terminal signal peptide, and targets the mature core protein. Consider that glycosylation may cause a higher apparent molecular weight in WB.
  • For WB, use appropriate loading controls and validate detection in serglycin-expressing cell lines (e.g., mast cells, T-cells). The predicted core protein weight is ~17.6 kDa, but observed bands may appear as broad smears.
  • In ELISA, this antibody can be used for detection; pairing with a capture antibody may enable sandwich ELISA development. Optimize coating and blocking conditions for your specific assay.
  • Cross-reactivity with mouse serglycin is reported; ensure sample compatibility and validate in the relevant species context.
