Biological Background: SRF Function and Localization
- SRF (Serum Response Factor), also designated MCM1, is a ubiquitously expressed transcription factor that binds to the serum response element (SRE) in the promoter region of immediate-early genes such as FOS.
- As a master regulator of the actin cytoskeleton, SRF forms a complex with the myocardin-related transcription factor A (MRTFA) to drive expression of cytoskeletal genes in response to Rho GTPase signaling and changes in globular actin (G-actin) levels.
- SRF is essential for cardiac development and myogenic differentiation, playing a critical role in cardiomyocyte maturation and contractile gene expression.
- The protein localizes predominantly to the nucleus and is subject to multiple post-translational modifications including phosphorylation and glycosylation, which modulate its transcriptional activity.
- SRF is classified as a developmental protein and a transcriptional activator with a conserved DNA-binding domain that recognizes the CC(A/T)6GG (CArG) box motif.
Experimental Guidance and Technical Tips
- For Western blot, a band at approximately 52 kDa may be observed in nuclear extracts of human or mouse cell lines; consider using positive controls such as HeLa or NIH/3T3 cells.
- The immunogen spans the N-terminal 280 amino acids, which includes the highly conserved DNA-binding domain, supporting cross-reactivity with mouse; validate antibody performance in your specific sample system.
- For ELISA, the antibody can be employed as a capture or detection reagent; optimal dilutions and buffer conditions should be empirically determined for each assay.
