Biological Background: SREBF2 Function and Localization

  • SREBF2 (Sterol Regulatory Element Binding Transcription Factor 2) is a basic helix-loop-helix leucine zipper (bHLH-LZ) transcription factor critical for lipid homeostasis.
  • It acts as a master transcription factor for cholesterol biosynthesis and uptake, regulating genes like HMG-CoA reductase and LDL receptor.
  • SREBF2 is synthesized as an inactive precursor anchored in the endoplasmic reticulum (ER) membrane. Related references: PMID:32322062
  • Under low cholesterol levels, the precursor is transported to the Golgi and proteolytically processed to release the active N-terminal domain, which translocates to the nucleus to activate target gene expression. Related references: PMID:32322062
  • It binds with high affinity to the sterol regulatory element 1 (SRE-1; 5′-ATCACCCCAC-3′) and also recognizes E-box motifs (5′-ATCACGTGA-3′), enabling regulation of diverse promoters. Related references: PMID:12177166 PMID:7903453
  • SREBF2 is ubiquitously expressed across adult and fetal human tissues, reflecting its fundamental role in cellular cholesterol metabolism.
  • The protein shuttles between the ER, Golgi, COPII vesicles, and nucleus, with subcellular distribution tightly controlled by cellular sterol status.
  • Post-translational modifications including phosphorylation, ubiquitination, and isopeptide bond formation modulate its stability and activity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 401–500 within the N-terminal transcription factor domain; both the full-length precursor (~124 kDa) and the cleaved nuclear form (~68 kDa) may be detected by Western blot depending on cell treatment conditions.
  • For Western blots, consider using protease inhibitors and performing sterol depletion (e.g., with statins) to enrich the nuclear active form.
  • In immunofluorescence experiments, permeabilization and staining of cells cultured under low-sterol conditions can enhance nuclear signal localization.
  • Cross-reactivity with mouse SREBF2 is reported; however, validate the antibody in your specific mouse tissue or cell line, as expression levels can vary.
  • The antibody is suitable for ELISA applications; titrate for optimal signal-to-noise ratio and include appropriate controls.
