Biological Background: SREBF1 Function and Localization

  • SREBF1 (also known as SREBP1, bHLHd1) encodes a sterol regulatory element-binding transcription factor that exists as a precursor embedded in the endoplasmic reticulum membrane. Upon low sterol concentrations, it is processed to release the transcription factor form that translocates to the nucleus. Related references: PMID:32322062
  • Cleaved SREBF1 binds to sterol regulatory element 1 (SRE-1; 5'-ATCACCCCAC-3') and E-box motifs (5'-ATCACGTGA-3') in the promoters of genes involved in cholesterol biosynthesis and the LDL receptor pathway, thereby activating their transcription.   • Alternative splicing produces isoforms with distinct activities: the SREBP-1A isoform (expressed in select tissues) has high transcriptional activity and stimulates both lipogenic and cholesterogenic gene expression, whereas the predominant SREBP-1C isoform (expressed in liver, adipose, and other tissues) primarily regulates lipogenic genes and has weaker activity.   • Additionally, an isoform lacking the requirement for Golgi proteolytic processing is constitutively active in transactivating lipogenic gene promoters.

  • In Western blot experiments, expect a band at approximately 122 kDa corresponding to the full-length precursor; a smaller band (~68 kDa) representing the cleaved active transcription factor may also be observed depending on cellular sterol status.
  • For immunofluorescence, localization may include cytoplasmic/endoplasmic reticulum patterns under sterol-replete conditions and nuclear translocation upon sterol depletion; co-staining with organelle markers is recommended.
