Biological Background: SR Protein Repeat (SRSF6/SRP55) Function and Localization
- Also known as Pre-mRNA-splicing factor SRP55, Splicing factor arginine/serine-rich 6 (SRSF6), and B52.
- Functions in constitutive pre-mRNA splicing and regulates alternative splice site selection.
- Modulates alternative splicing of MAPT/Tau exon 10, thereby influencing tau protein isoform expression.
- Binds to alternative exons of tenascin-C (TNC) pre-mRNA and promotes the expression of alternatively spliced TNC variants.
- Involved in wound healing and keratinocyte differentiation/proliferation through its role in alternative splicing regulation.
- Localizes to the nucleus and accumulates in nuclear speckles, which are sites of active splicing and mRNA processing.
- Post-translational modifications include phosphorylation, acetylation, and isopeptide bond formation (ubiquitination), which may regulate its activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 100–200 of human SRSF6; high sequence conservation with mouse and rat supports the observed cross-reactivity.
- For western blot, a band of approximately 40 kDa is expected; use appropriate nuclear loading controls and validate in relevant cell lines or tissues.
- IHC-P may require antigen retrieval; confirm staining patterns in tissues known to express SRSF6, such as those with active splicing.
- IP and ELISA can be optimized by titrating antibody concentration and using appropriate positive controls.
