Biological Background: SQSTM1/p62 Function and Localization
- SQSTM1/p62 (Sequestosome-1), also known as p62, EBIAP, p60, and ubiquitin-binding protein p62, is a multifunctional scaffold protein involved in selective autophagy, signaling, and cellular stress responses.
- Core function as an autophagy receptor: SQSTM1 bridges polyubiquitinated proteins and autophagosomes. It undergoes liquid-liquid phase separation upon binding ubiquitinated cargo via its UBA domain, forming p62 bodies, and then engages ATG8 family proteins on autophagosomal membranes through its LIR motif, leading to degradation of both cargo and SQSTM1 itself. - For Western blot (WB), expect a band at approximately 48 kDa. SQSTM1 is degraded by autophagy; treatment with lysosomal inhibitors (e.g., chloroquine or bafilomycin A1) can increase detection levels.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval conditions and validate with appropriate controls, as SQSTM1 forms distinct cytoplasmic puncta (p62 bodies) that are enriched under stress conditions.
- Cross-reactivity is reported for human, mouse, and rat; however, it is advisable to verify performance in your specific sample system if using non-human species.
