- Protein sprouty homolog 2 (SPRY2) is a member of the sprouty family that antagonizes receptor tyrosine kinase signaling, particularly the fibroblast growth factor (FGF) pathway.
- SPRY2 inhibits FGF-mediated phosphorylation of ERK1/2, thereby acting as a negative regulator of FGF-induced retinal lens fiber differentiation, limb bud outgrowth, and respiratory organogenesis (By similarity).
- It also inhibits transforming growth factor beta (TGFB)-induced epithelial-to-mesenchymal transition in retinal lens epithelial cells (By similarity).
- SPRY2 inhibits CBL/C-CBL-mediated EGFR ubiquitination, affecting receptor downregulation. Related references: PMID:17974561
- Subcellularly, SPRY2 localizes to the cytoplasm, cytoskeleton, cell projections, and ruffle membranes, indicating roles in cell dynamics and signaling.
