Biological Background: SPINT1 (HAI-1) Function and Localization
- SPINT1 (also known as HAI-1) is a Kunitz-type serine protease inhibitor that inhibits hepatocyte growth factor activator (HGFAC) (PubMed:9045658). Related references: PMID:9045658
- It inhibits the serine protease activity of ST14/matriptase in vitro (PubMed:28710277). Related references: PMID:28710277
- SPINT1 inhibits TMPRSS13 serine protease activity via its BPTI/Kunitz inhibitor 1 domain (PubMed:20977675). Related references: PMID:20977675
- The protein localizes to the secreted compartment, cytoplasm, and cell membrane (UniProt: Secreted, Cytoplasm, Cell membrane).
- It is a glycoprotein containing disulfide bonds (UniProt KEYWORDS: Glycoprotein, Disulfide bond) and features two Kunitz-type inhibitor domains (BPTI/Kunitz).
- Alternative splicing produces multiple isoforms (UniProt KEYWORDS: Alternative splicing).
- SPINT1 is involved in regulating proteolysis and cellular signaling pathways through its inhibitory functions.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 234-513 of human SPINT1, covering the C-terminal region possibly including the Kunitz domains and transmembrane segment; this design may enhance epitope accessibility.
- For western blot (WB), the predicted molecular weight is 58 kDa; however, due to glycosylation and other post-translational modifications, the apparent molecular weight may vary. Consider using appropriate protein ladders and optimizing transfer conditions.
- The antibody is reported to cross-react with mouse and rat SPINT1; thus, it may be suitable for studies in rodent models, but validation in your specific sample system is recommended.
- For immunofluorescence (IF/ICC), consider permeabilization protocols that allow detection of both membrane and cytoplasmic pools.
- For ELISA, the antibody can be used as a capture or detection reagent; perform checkerboard titrations to determine optimal concentrations.
