Biological Background: SPINK5/LEKTI (Serine Protease Inhibitor Kazal-type 5) Function and Localization

  • SPINK5 (Serine Peptidase Inhibitor Kazal Type 5) encodes the serine protease inhibitor LEKTI (Lympho-Epithelial Kazal-Type-related Inhibitor), a secreted multidomain protein belonging to the Kazal-type serine protease inhibitor family. It contains multiple Kazal domains and disulfide bonds, with a molecular weight of approximately 121 kDa.
  • LEKTI functions as a key regulator of protease activity in the skin and mucous epithelia, inhibiting kallikreins (KLK5, KLK7, KLK14), caspase-14 (CASP14), and trypsin in a pH-dependent manner. This inhibition is crucial for maintaining the protective barrier of the skin and controlling desquamation and inflammation.
  • The protein is highly expressed in the thymus and the stratum corneum of the skin, with additional detection in the oral mucosa, parathyroid gland, Bartholin's glands, tonsils, and vaginal epithelium. It is found at very low levels in lung, kidney, and prostate.
  • LEKTI is a secreted protein that undergoes proteolytic processing, including cleavage at paired basic residues, generating multiple active fragments. Its alternative splicing produces distinct isoforms.
  • Due to its role in inhibiting pro-inflammatory and pro-desquamatory proteases, LEKTI contributes to antimicrobial defense and prevents excessive tissue breakdown; mutations in SPINK5 are associated with Netherton syndrome, characterized by severe skin barrier defects.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 100–200 of human SPINK5/LEKTI (Q9NQ38), located within the N-terminal region. For Western blot (WB), consider using whole-cell lysates from tissues with high LEKTI expression (e.g., skin, thymus) or recombinant protein controls to validate antibody specificity. The predicted molecular weight is approximately 121 kDa, but processed fragments may also be detected.
  • For ELISA applications, this monoclonal antibody may be used as a capture or detection reagent for quantifying LEKTI levels in biological fluids or tissue extracts. Ensure proper optimization of coating concentrations and blocking conditions.
  • Cross-reactivity with mouse and rat indicates conservation of the epitope, but sequence alignment of the immunogen region across species is recommended to confirm potential binding. Validate the antibody in the relevant sample system for your experimental setup.
  • Given the pH-dependent activity of LEKTI, consider the effect of pH on antibody–antigen binding if developing assays under different buffer conditions.
