Biological Background: SPINK1 Function and Localization
- SPINK1, also known as pancreatic secretory trypsin inhibitor (PSTI) or tumor-associated trypsin inhibitor (TATI), is a small secreted serine protease inhibitor of the Kazal-type 1 family.
- It exhibits anti-trypsin activity, which is critical for protecting the pancreas against trypsin-catalyzed premature activation of zymogens. Related references: PMID:7142173
- In the male reproductive tract, SPINK1 binds to sperm heads and modulates sperm capacitance by inhibiting calcium uptake and nitric oxide production.
- The protein is secreted and contains three conserved disulfide bonds, characteristic of Kazal-type inhibitors.
- Clinically, mutations in SPINK1 are linked to hereditary pancreatitis, making it a key target in pancreatic disease research.
Experimental Guidance and Technical Tips
- For IF/ICC, consider using SPINK1-expressing cell lines such as pancreatic acinar cells or sperm cells, with appropriate fixation and permeabilization. Validate staining specificity with known positive/negative controls.
- For ELISA, pair with a matched detection antibody or use as a capture/detection antibody, optimizing coating conditions and sample dilution. The recombinant immunogen (aa 24-79) may help assess sensitivity.
- The antibody may recognize both native and denatured SPINK1 due to the recombinant immunogen; however, additional validation under experimental conditions is recommended.
