Biological Background: SPIN1 Function and Localization
- Encoded by the SPIN1 gene, Spindlin-1 is a chromatin reader protein also known as Ovarian cancer-related protein, belonging to the SPIN family.
- Specifically recognizes and binds histone H3 trimethylated at both Lys-4 and Lys-9 (H3K4me3K9me3), playing a critical role in piRNA-mediated retrotransposon silencing during spermatogenesis. Related references: PMID:33574238
- Key initiator of the PIWIL4-piRNA pathway in male embryonic germ cells, promoting DNA methylation and silencing of young LINE1 transposons by recruiting PIWIL4 and SPOCD1.
- Also recognizes H3K4me3 and H3R8me2a, acting as an activator of the Wnt signaling pathway downstream of PRMT2. Related references: PMID:22258766, PMID:29061846
- In cancer, promotes cell proliferation via Wnt signaling activation; overexpression induces metaphase arrest and chromosomal instability. Related references: PMID:24589551
- Localizes to the nucleus and nucleolus, where it associates with active rDNA loci and promotes rRNA gene expression. Related references: PMID:21960006
- May participate in cell-cycle regulation during the gamete-to-embryo transition and in oocyte meiotic resumption.
- Highly expressed in ovarian cancer tissues; post-translationally modified by acetylation, phosphorylation, and isopeptide bond formation, with functional roles in meiosis and chromatin regulation.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 1–262 of human SPIN1, suggesting recognition of the full-length protein; consider that native conformations may be detected.
- For Western blot, a band near 30 kDa is expected; validate with appropriate loading controls and confirm band size in your sample system.
- For IHC-P, antigen retrieval conditions (e.g., heat-induced epitope retrieval at pH 6 or 9) should be optimized based on tissue type and fixation method.
- For ELISA, the antibody can be used as a capture or detection reagent; perform checkerboard titrations to determine optimal dilutions.
- Cross-reactivity with mouse and rat indicates conserved epitopes; validate reactivity in the relevant species’ tissue or cell lysates.
- Due to the nuclear localization of SPIN1, ensure efficient nuclear protein extraction for Western blot or appropriate permeabilization for immunostaining.
- Multiple post‑translational modifications (acetylation, phosphorylation, ubiquitination‑like) may affect antibody binding; consider phosphatase or deacetylase treatment if band shifts are observed.
