Biological Background: SPI1 Function and Localization
- Pioneers hematopoietic transcription by decompacting stem cell heterochromatin, enabling access for other transcription factors, and directly controlling gene expression through genetic regulatory elements (PubMed:23658224, PubMed:33951726). Related references: PMID:23658224, PMID:33951726
- Binds via its ETS domain to the purine-rich DNA core sequence 5'-GAGGAA-3' (PU-box) to regulate target genes (PubMed:33951726). Related references: PMID:33951726
- Transactivates genes critical for myeloid and lymphoid lineage commitment, such as CSF1R.
- In vitro, can also bind RNA and interfere with pre-mRNA splicing.
- Localizes to the nucleus to execute transcriptional regulatory functions.
- Highly expressed in bone marrow hematopoietic stem cells, lymphoid progenitors, myeloid lineages, and pre-B1 cells.
- Also found in germinal center B-cells.

- The immunogen spans amino acids 17-98 of human SPI1, targeting the N-terminal region; this may allow detection of full-length SPI1.
- For Western blot, a specific band around 31 kDa is expected; use appropriate positive and negative controls to validate reactivity.
