Biological Background: SPHK2 Function and Localization
- Catalyzes the phosphorylation of sphingosine to form sphingosine-1-phosphate (SPP), a lipid mediator with intra- and extracellular functions; also acts on other sphingoid bases and binds phosphoinositides. Related references: PMID:12954646, PMID:19168031
- Unlike prosurvival SPHK1, SPHK2 positively regulates intracellular ceramide levels, inhibits cell growth, and enhances apoptosis. Related references: PMID:16118219
- In mitochondria, SPHK2-produced SPP binds PHB2 to modulate cytochrome-c oxidase assembly and mitochondrial respiration. Related references: PMID:20959514
- In the nucleus, SPHK2 interacts with HDAC1/HDAC2, inhibiting their deacetylase activity via SPP, leading to increased acetylation of histones H3-K9, H4-K5, and H2B-K12, thereby regulating epigenetic gene expression. Related references: PMID:19729656
- Nuclear SPHK2 may also inhibit DNA synthesis and cell cycle progression. Related references: PMID:12954646, PMID:16103110
- Localizes to multiple cellular compartments, including cytoplasm, nucleus, endoplasmic reticulum, mitochondrial inner membrane, and lysosome membrane.
- Predominantly expressed in kidney, liver, and brain, with isoform 1 being the major form in most tissues; detected in cerebral cortex and hippocampus at the protein level. Related references: PMID:10751414, PMID:29615132, PMID:16103110
- Annotated as an ATP-binding kinase involved in lipid metabolism; features alternative splicing and phosphorylation modifications.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (aa 1-165) of human SPHK2, a domain conserved across isoforms; the antibody may recognize multiple splice variants.
- For Western blot (WB), consider using lysates from cell lines or tissues known to express SPHK2, such as HEK293, HeLa, or kidney/liver/brain samples, with appropriate loading controls.
- In ELISA, optimize the coating antibody concentration and buffer conditions according to your plate format and detection system; include a standard curve for quantification.
- Because SPHK2 associates with membranes, include mild detergents in lysis buffers to ensure efficient extraction and detection.
