Biological Background: SPHK2 Function and Localization

  • Catalyzes the phosphorylation of sphingosine to form sphingosine-1-phosphate (SPP), a lipid mediator with intra- and extracellular functions; also acts on other sphingoid bases and binds phosphoinositides. Related references: PMID:12954646, PMID:19168031
  • Unlike prosurvival SPHK1, SPHK2 positively regulates intracellular ceramide levels, inhibits cell growth, and enhances apoptosis. Related references: PMID:16118219
  • In mitochondria, SPHK2-produced SPP binds PHB2 to modulate cytochrome-c oxidase assembly and mitochondrial respiration. Related references: PMID:20959514
