Biological Background: SPHK2 Function and Localization

  • SPHK2 (SK2) is a sphingosine kinase that catalyzes the phosphorylation of sphingosine to sphingosine-1-phosphate (S1P). It also acts on D-erythro-dihydrosphingosine, D-erythro-sphingosine and L-threo-dihydrosphingosine, and binds phosphoinositides. Related references: PMID:12954646, PMID:19168031
  • Unlike prosurvival SPHK1, SPHK2 positively regulates intracellular ceramide levels, inhibits cell growth, and enhances apoptosis. Related references: PMID:16118219
  • In mitochondria, SPHK2-generated S1P binds PHB2 to modulate cytochrome-c oxidase assembly and mitochondrial respiration through complex IV. Related references: PMID:20959514
  • Nuclear SPHK2 interacts with HDAC1 and HDAC2; S1P production inhibits their deacetylase activity, leading to increased acetylation of histones H3-K9, H4-K5, and H2B-K12, thereby regulating epigenetic gene expression. Related references: PMID:19729656
  • In the nucleus, SPHK2 may suppress DNA synthesis and cell cycle progression. Related references: PMID:12954646, PMID:16103110
  • SPHK2 localizes to multiple subcellular compartments including cytoplasm, nucleus, endoplasmic reticulum, mitochondrial inner membrane, and lysosome membrane.
  • It is highly expressed in adult kidney, liver, and brain, with enrichment in cerebral cortex and hippocampus; isoform 1 predominates in most tissues. Related references: PMID:10751414, PMID:29615132, PMID:16103110

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1-165, an N-terminal region; consider that this antibody may detect full-length SPHK2 (~69kDa) and possibly N-terminal-containing isoforms. Validate specificity in the relevant sample system.
  • For Western blotting, human kidney, liver, or brain lysates are recommended positive controls given the high expression pattern; mouse samples may also be tested to confirm cross-reactivity.
  • Given the multiple subcellular localizations of SPHK2, consider appropriate subcellular fractionation or treatment conditions when analyzing protein distribution.
  • For ELISA applications, consider optimization of the antibody pair and assay conditions; this polyclonal can be used as either capture or detection reagent in a sandwich ELISA format with proper validation.
