Biological Background: SPHK2 Function and Localization
- SPHK2 (SK2) is a sphingosine kinase that catalyzes the phosphorylation of sphingosine to sphingosine-1-phosphate (S1P). It also acts on D-erythro-dihydrosphingosine, D-erythro-sphingosine and L-threo-dihydrosphingosine, and binds phosphoinositides. Related references: PMID:12954646, PMID:19168031
- Unlike prosurvival SPHK1, SPHK2 positively regulates intracellular ceramide levels, inhibits cell growth, and enhances apoptosis. Related references: PMID:16118219
- In mitochondria, SPHK2-generated S1P binds PHB2 to modulate cytochrome-c oxidase assembly and mitochondrial respiration through complex IV. Related references: PMID:20959514
