Biological Background: SPHK1 Function and Localization
- SPHK1 (sphingosine kinase 1), also known as SK1 and acetyltransferase SPHK1, is a multifunctional enzyme encoded by the SPHK1 gene (Gene ID: 8877) in humans. It belongs to the sphingosine kinase family and plays key roles in sphingolipid metabolism and signaling.
- Catalyzes the phosphorylation of sphingosine to form sphingosine 1-phosphate (S1P), a bioactive lipid mediator with intracellular and extracellular functions. - In contrast to proapoptotic SPHK2, negatively regulates intracellular ceramide levels, enhances cell growth, and inhibits apoptosis. Related references: PMID:16118219

- The immunogen was designed against the N-terminal region (aa 1-160) of human SPHK1 (NP_001136074.1). This region is common to multiple isoforms and may provide broad detection; verify sequence alignment for isoform-specific needs.
- For Western blot, a band near 43 kDa is expected, but post-translational modifications or alternative splicing may result in band shifts. Optimize loading controls and antibody dilution using relevant positive and negative lysates.
- For ELISA, consider pairing with capture/detection antibodies validated for SPHK1. Validate assay specificity using recombinant protein and appropriate negative controls.
