- SPDEF (Prostate-derived Ets factor), also known as PDEF, is a SAM pointed domain-containing Ets transcription factor encoded by the SPDEF gene. It belongs to the ETS family of transcription factors and contains a pointed (PNT) domain and an ETS DNA-binding domain.
- SPDEF functions as an androgen-independent transactivator of the prostate-specific antigen (PSA) promoter and binds specifically to 5'-GGAT-3' DNA sequences. This activity highlights its role in prostate epithelial regulation.
- It also acts as a transcriptional activator for the SERPINB5 (maspin) promoter, a gene implicated in tumor suppression and inhibition of metastasis.
- Tissue expression is highly restricted to hormone-regulated epithelial tissues, with the highest levels in the prostate gland. Significantly lower expression is observed in mammary gland, salivary gland, and ovary. SPDEF is also expressed in prostate carcinoma cells.
