  • SPDEF (Prostate-derived Ets factor), also known as PDEF, is a SAM pointed domain-containing Ets transcription factor encoded by the SPDEF gene. It belongs to the ETS family of transcription factors and contains a pointed (PNT) domain and an ETS DNA-binding domain.
  • SPDEF functions as an androgen-independent transactivator of the prostate-specific antigen (PSA) promoter and binds specifically to 5'-GGAT-3' DNA sequences. This activity highlights its role in prostate epithelial regulation.
  • It also acts as a transcriptional activator for the SERPINB5 (maspin) promoter, a gene implicated in tumor suppression and inhibition of metastasis.
