Biological Background: SPAG8 Function and Localization
- SPAG8, also known as SMP-1, BS-84, HSD-1, or CT142, is a sperm-associated antigen and a microtubule inner protein (MIP) component of the dynein-decorated doublet microtubules (DMTs) in cilia and flagella axonemes.
- SPAG8 is required for motile cilia beating, as demonstrated in airway epithelial cells. Related references: PMID:36191189
- In spermatogenesis, SPAG8 enhances the binding of CREM isoform tau to its coactivator FHL5, increasing FHL5-regulated transcriptional activation (By similarity).
- SPAG8 participates in the acrosome reaction and the binding of sperm to the zona pellucida during fertilization (By similarity).
- The protein regulates the cell cycle by controlling progression through the G2/M phase, possibly by delaying CDK1 activation required for mitotic entry. Related references: PMID:19548270
- SPAG8 interacts with RANBP9, a connection that may influence microtubule formation and fertility. Related references: PMID:10500252
- Subcellular localization includes the cytoplasm, nucleus, acrosome, spindle, and the axonemes of cilia and flagella, reflecting its diverse functional roles.
- Expression is predominantly in testis germ cells and airway epithelial cells; it is absent from tissues such as liver, kidney, prostate, and small intestine.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 220–420 of human SPAG8, a region that may capture multiple isoforms; consider epitope mapping if isoform specificity is required.
- For Western blot, the predicted molecular weight is approximately 51 kDa; include positive control lysates from testis or ciliated cell lines (e.g., airway epithelial cells) to confirm reactivity.
- Given cross-reactivity with mouse and rat, the antibody can be used for comparative studies; verify species-specific background levels in your model system.
- In ELISA applications, validate detection against the recombinant immunogen or native SPAG8 from relevant biological fluids/extracts.
- As a polyclonal antibody, lot-to-lot consistency should be assessed; optimize working concentrations for each experimental setup.
