Biological Background: SPAG5 Function and Localization
- SPAG5 (Sperm-associated antigen 5), also known as Astrin, Deepest, and MAP126, is a 134 kDa protein highly expressed in testis. It localizes to the cytoplasm, mitotic spindle, spindle poles, kinetochores, midbody, and centrosomes.
- Essential component of the mitotic spindle required for normal chromosome segregation and progression into anaphase. Related references: PMID:11724960, PMID:12356910, PMID:27462074
- Required for chromosome alignment, normal timing of sister chromatid segregation, and maintenance of spindle pole architecture. Related references: PMID:17664331, PMID:27462074
- In complex with SKAP, promotes stable microtubule-kinetochore attachments and may regulate separase activity.
- May regulate AURKA localization to mitotic spindle and CCNB1 localization to both spindle and centrosomes. Related references: PMID:18361916, PMID:21402792
- Involved in centriole duplication; required for centrosomal localization of CDK5RAP2, CEP152, WDR62, and CEP63, and promotes centrosomal localization of CDK2. Related references: PMID:26297806
- In non-mitotic cells, inhibits mTORC1 by recruiting RPTOR to stress granules upon stress, preventing mTORC1 hyperactivation-induced apoptosis. Related references: PMID:23953116
- May enhance GSK3B-mediated phosphorylation of substrates such as MAPT/TAU. Related references: PMID:18055457
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering amino acids 1-300 of human SPAG5, which may generate antibodies targeting multiple epitopes along the N-terminal region.
- For immunofluorescence (IF/ICC), consider using mitotic cells (e.g., HeLa or U2OS synchronized with nocodazole) to visualize spindle and kinetochore localization. Optimize fixation/permeabilization protocols to preserve spindle structure.
- For ELISA, recombinant SPAG5 protein or cell lysates from testis or mitotically active cells can serve as positive controls. Perform antibody titration to determine the optimal working dilution.
- Validate antibody specificity in the relevant sample system (e.g., siRNA knockdown or CRISPR knockout) to confirm specific detection of SPAG5.
