Biological Background: SP100 Function and Localization
- SP100 is a nuclear autoantigen and tumor suppressor, a major constituent of PML nuclear bodies, involved in cell growth, differentiation, and apoptosis.
- It functions as a transcriptional coactivator of ETS1 and ETS2, enhancing their transcriptional activity, but can also act as a corepressor of ETS1 under certain conditions, inhibiting its DNA binding. Related references: PMID:11909962, PMID:15247905
- SP100 plays a role in angiogenesis by regulating endothelial cell motility and invasion through ETS1.
- It interacts with the MRN complex to regulate telomere lengthening.
- SP100 may regulate TP53-mediated transcription and, through CASP8AP2, regulate FAS-mediated apoptosis.
- The protein localizes primarily to the nucleus, specifically to PML bodies and nuclear bodies, but can also be found in the cytoplasm.
