Biological Background: SP100 Function and Localization

  • SP100 is a nuclear autoantigen and tumor suppressor, a major constituent of PML nuclear bodies, involved in cell growth, differentiation, and apoptosis.
  • It functions as a transcriptional coactivator of ETS1 and ETS2, enhancing their transcriptional activity, but can also act as a corepressor of ETS1 under certain conditions, inhibiting its DNA binding. Related references: PMID:11909962, PMID:15247905
  • SP100 plays a role in angiogenesis by regulating endothelial cell motility and invasion through ETS1.
  • It interacts with the MRN complex to regulate telomere lengthening.
  • SP100 may regulate TP53-mediated transcription and, through CASP8AP2, regulate FAS-mediated apoptosis.
  • The protein localizes primarily to the nucleus, specifically to PML bodies and nuclear bodies, but can also be found in the cytoplasm.
  • Widely expressed tissues, but isoform Sp100-B is restricted to spleen, tonsil, thymus, and certain B and T cell lines.
  • Post-translational modifications include acetylation, phosphorylation, and isopeptide bond formation; it also features alternative splicing and a coiled-coil domain.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 181-480 of human SP100, a central region that may be recognized by the polyclonal antibody in both human and mouse samples due to sequence conservation.
  • For Western blot, expect bands around 100 kDa; however, alternative splicing isoforms may produce additional bands; validate with a known positive control such as HeLa cell lysate.
  • In ELISA applications, this antibody can be used to detect recombinant or native SP100; optimize coating and detection conditions using the recombinant protein immunogen.
  • Since SP100 is a nuclear protein, ensure proper nuclear extraction protocols for effective detection in WB.
  • Cross-reactivity with mouse is reported; verify sequence homology of the immunogen region between human and mouse SP100 for quantitative applications.
