Biological Background: SOX9 Function and Localization
- Transcription factor SOX-9 is a member of the SOX (SRY-related HMG-box) family of transcription factors, encoded by the SOX9 gene in humans (Gene ID: 6662). Alternative names include CMD1, SRA1, CMPD1, SRXX2, and SRXY10.
- SOX9 plays a critical role in chondrocyte differentiation and skeletal development, specifically binding the 5'-ACAAAG-3' DNA motif in enhancers and super-enhancers to drive expression of cartilage matrix genes like COL2A1, COL4A2, COL9A1, COL11A2, ACAN, SOX5, and SOX6 (PubMed:8640233). - It is absolutely required for precartilaginous condensation, the first step in chondrogenesis, and together with SOX5 and SOX6, drives overt chondrogenesis and subsequent chondrocyte maturation.
