- Transcription factor SOX-6 (SRY-box 6) is a member of the SOX family of transcription factors, characterized by a conserved HMG-box DNA-binding domain. It plays critical roles in development, particularly in chondrogenesis and neurogenesis.
- SOX6 specifically binds the DNA motif 5'-AACAAT-3' present in enhancers and super-enhancers, thereby promoting expression of genes crucial for cartilage formation. It cooperates with SOX5 and SOX9 to transactivate cartilage-specific genes.
- During chondrogenesis, SOX6 is essential for the overt differentiation of prechondrocytes into early stage chondrocytes, but it is not required for the initial precartilaginous condensation step.
- In the developing spinal cord, SOX6 is involved in oligodendrocyte differentiation; it binds to the proximal promoter of the myelin protein MPZ gene and acts as a transcriptional repressor of MBP.
- SOX6 localizes to both the nucleus and cytoplasm. Its activity is regulated by post-translational modifications including phosphorylation, ubiquitination, and isopeptide bond formation.
- The protein is widely expressed, with highest levels in skeletal muscle. Alternative splicing generates multiple isoforms. Mutations or altered expression of SOX6 have been associated with autism spectrum disorder and intellectual disability.
