• Transcription factor SOX-6 (SRY-box 6) is a member of the SOX family of transcription factors, characterized by a conserved HMG-box DNA-binding domain. It plays critical roles in development, particularly in chondrogenesis and neurogenesis.
• SOX6 specifically binds the DNA motif 5'-AACAAT-3' present in enhancers and super-enhancers, thereby promoting expression of genes crucial for cartilage formation. It cooperates with SOX5 and SOX9 to transactivate cartilage-specific genes.
• During chondrogenesis, SOX6 is essential for the overt differentiation of prechondrocytes into early stage chondrocytes, but it is not required for the initial precartilaginous condensation step.
