Biological Background: SOX4 Function and Localization
- Transcription factor SOX-4 is a member of the SOX (SRY-related HMG-box) family of DNA-binding proteins, acting as a transcriptional activator.
- Binds with high affinity to the T-cell enhancer motif 5'-AACAAAG-3', implicating it in immune cell gene regulation. Related references: PMID:30661772
- Plays a critical role in the maturation and development of IL17A-producing Vγ2-positive γδ T-cells by modulating RORC expression (By similarity).
- Involved in skeletal myoblast differentiation by promoting CALD1 gene expression. Related references: PMID:26291311
- Localizes exclusively to the nucleus, consistent with its function as a transcription factor.
- Predominantly expressed in testis, brain, and heart, suggesting roles in neural and cardiac development.
- Post-translational modifications include acetylation, which may regulate its transcriptional activity.
Experimental Guidance and Technical Tips
- For Western blot, an expected band near 47 kDa may be observed. As a polyclonal antibody, lot-to-lot consistency should be titrated and validated in the relevant sample system.
- Given the immunogen's N-terminal peptide sequence (aa 1–100), the antibody may recognize full-length SOX4. Cross-reactivity with mouse suggests conservation of this epitope; consider this for interspecies studies.
- In ELISA, the antibody may be suitable for detecting recombinant or endogenous SOX4; pairing with a monoclonal antibody could improve specificity.
- Target expression is highest in testis, brain, and heart; these tissues could serve as positive controls, but expression levels should be verified under your experimental conditions.
