Biological Background: SOX2 Function and Localization
- Transcription factor SOX2 (SRY-box transcription factor 2) forms a trimeric complex with OCT4 on DNA, controlling the expression of genes such as YES1, FGF4, UTF1, and ZFP206 involved in embryonic development (By similarity).
- It binds to the proximal enhancer region of NANOG and is critical for early embryogenesis and embryonic stem cell pluripotency. Related references: PMID:18035408
- As a downstream target of SRRT, SOX2 promotes neural stem cell self-renewal and maintains neural cells in an undifferentiated state by counteracting the activity of proneural proteins and suppressing neuronal differentiation (By similarity).
- The protein localizes to the nucleus, nuclear speckles, and cytoplasm.
- Post-translational modifications include phosphorylation, methylation, isopeptide bond formation, and ubiquitin-like conjugation.
- SOX2 is a developmental protein and transcriptional activator, involved in transcription regulation and DNA binding.
- Mutations in SOX2 are associated with congenital anomalies including anophthalmia and microphthalmia.
Experimental Guidance and Technical Tips
- The immunogen (aa 1–140 of human SOX2) targets the N-terminal region; consider that this may detect full-length as well as N-terminal fragments or isoforms.
- For western blotting, the predicted molecular weight is 34 kDa; optimize gel percentage and transfer conditions accordingly.
- In immunohistochemistry (IHC) and immunofluorescence (IF), validate antigen retrieval methods and include appropriate controls, noting that SOX2 can localize to the nucleus and cytoplasm.
- For immunoprecipitation (IP), ensure antibody compatibility with your lysis buffer and consider that SOX2 may be in complex with other proteins.
- Cross-reactivity with human, mouse, and rat has been reported; validate in your specific tissue or cell type.
- Determine optimal working dilution empirically for each application.
