Biological Background: SOX2 Function and Localization
- SOX2 is a transcription factor that forms a trimeric complex with OCT4 on DNA, controlling expression of genes like YES1, FGF4, UTF1, and ZFP206 involved in embryonic development.
- It binds to the proximal enhancer region of NANOG, a key pluripotency factor, and is critical for early embryogenesis and embryonic stem cell pluripotency. Related references: PMID:18035408
- SOX2 acts downstream of SRRT to promote neural stem cell self-renewal and keeps neural cells undifferentiated by counteracting proneural proteins, suppressing neuronal differentiation.
- It may function as a developmental switch in neuronal differentiation.
- Alternative names for SOX2 include ANOP3 and MCOPS3, associated with microphthalmia and other developmental disorders (disease variant keyword).
- Subcellular localization includes nucleus speckles, cytoplasm, and nucleus, and the protein is subject to post-translational modifications such as phosphorylation, methylation, and ubiquitination (Ubl conjugation).
- The human SOX2 protein has a molecular weight of approximately 34 kDa (34310 Da from UniProt).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (aa 1-100) of human SOX2. For Western blot, a band around 34 kDa is expected; consider using positive controls like human embryonic stem cell lysates.
- For immunofluorescence (IF-F), fixation and permeabilization conditions may need optimization to detect nuclear-localized SOX2; validate in relevant cell lines.
- Cross-reactivity with mouse and rat has been reported; however, we recommend confirming species-specific reactivity in your experimental setup.
- For ELISA, pair with an appropriate capture/detection antibody and validate using recombinant SOX2 protein or cell lysates.
- Given the role of SOX2 in stem cell pluripotency, this antibody is suitable for developmental biology and stem cell research applications.
