Biological Background: SOX18 Function and Localization
- SOX18 is a member of the SOX (SRY-related HMG-box) family of transcription factors, characterized by a conserved HMG-box DNA-binding domain.
- It functions as a transcriptional activator, recognizing the consensus sequence 5'-AACAAAG-3' in target gene promoters.
- Essential for embryonic cardiovascular development and lymphangiogenesis; activates expression of PROX1 and other lymphatic endothelial markers.
- Triggers differentiation of lymphatic vessels, but is not required for the maintenance of differentiated lymphatic endothelial cells.
- Plays a functionally redundant role with SOX17 in postnatal angiogenesis.
- Interaction with MEF2C enhances its transcriptional activation activity.
- Required for normal hair development, and mutations are associated with hypotrichosis-lymphedema-telangiectasia syndrome (HLTS).
- Subcellularly localized to the nucleus, with tissue expression detected in heart, lung, placenta, skeletal muscle, liver, kidney, spleen, prostate, ovary, small intestine, and colon.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human SOX18; sequence alignment across species of interest should be performed to validate epitope conservation if cross-reactivity is critical.
- For WB, the predicted molecular weight is approximately 41 kDa; appropriate positive controls such as endothelial or lymphatic cell lysates may be considered.
- As a polyclonal antibody, lot-to-lot consistency should be verified in your experimental system, and specificity confirmed with appropriate negative controls.
- ELISA applications may require optimization of coating antigen concentration and antibody dilution to achieve the desired signal-to-noise ratio.
