Biological Background: Transcription factor SOX-17 Function and Localization
- SOX17 is a transcription factor of the SOX (SRY-related HMG-box) family, encoded by the SOX17 gene.
- Acts as a transcription regulator that binds to target promoter DNA sequences 5'-AACAAT-'3 or 5'-AACAAAG-3'. Related references: PMID:33952808
- Modulates WNT3A signaling and inhibits the Wnt pathway by promoting degradation of activated CTNNB1.
- Plays a key role in embryonic development, including definitive gut endoderm formation, heart tube looping, and vascular development.
- Required for fetal hematopoietic stem cell generation and maintenance, and acts as a probable transcriptional activator in premeiotic germ cells.
- Localizes to the nucleus.
- Expressed in adult heart, lung, spleen, testis, ovary, placenta, fetal lung and kidney; preferentially in esophagus, stomach and small intestine.
- Functionally redundant with SOX18 in postnatal angiogenesis.
- Associated with disease variant VUR3 (vesicoureteral reflux 3).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 343-414), which may provide specificity for SOX17 over other SOX family members.
- For WB, consider using nuclear extracts due to the nuclear localization of SOX17.
- Validate antibody performance in relevant cell lines or tissues expressing SOX17, such as those from heart, lung, or testis.
- Cross-reactivity with mouse and rat orthologs is expected; verify sequence homology in the immunogen region.
