Biological Background: SOX10 Function and Localization

  • SOX10 is a member of the SOX (SRY-related HMG-box) family of transcription factors, characterized by a conserved HMG DNA-binding domain.
  • It plays a central role in the development and maturation of glial cells, particularly oligodendrocytes, by activating myelin gene expression such as DUSP15 and MYRF.
  • Once induced, MYRF cooperates with SOX10 to implement the myelination program in the central nervous system.
  • SOX10 acts as a transcriptional activator of MITF, synergizing with PAX3 to regulate melanocyte development and differentiation. Related references: PMID:21965087
  • It directly binds to the promoter of the MBP (myelin basic protein) gene and activates its transcription.
  • Alternative splicing generates multiple isoforms; the full-length protein has a calculated molecular weight of about 50 kDa, while an isoform detected by this antibody runs at ~31 kDa.
  • Subcellular localization includes the nucleus, where it functions as a transcription factor, as well as the cytoplasm and mitochondrial outer membrane.
  • Tissue expression is detectable in fetal and adult brain, heart, small intestine, and colon.

Experimental Guidance and Technical Tips

  • For Western blot, use nuclear extracts from oligodendrocytes or melanoma cell lines as positive controls, anticipating bands at ~50 kDa and ~31 kDa.
  • In immunohistochemistry, antigen retrieval may be required; test on sections known to express SOX10 such as skin (melanocytes) or brain tissue.
  • This rabbit monoclonal antibody is suitable for immunoprecipitation; however, optimization of antibody concentration is recommended for each experimental system.
  • When performing immunofluorescence, verify colocalization with nuclear or mitochondrial markers, as SOX10 can localize to multiple subcellular compartments.
