Biological Background: SOSTDC1 Function and Localization
- SOSTDC1 (Sclerostin domain-containing protein 1), also known as Ectodin, USAG-1 (uterine sensitization-associated gene 1 protein), DAND7, and CDA019, is a secreted protein belonging to the sclerostin family. It is expressed as a glycoprotein containing a cystine-knot motif and intramolecular disulfide bonds.
- It functions as a direct antagonist of bone morphogenetic proteins BMP2, BMP4, BMP6, and BMP7 in a dose-dependent manner, thereby modulating BMP signaling.
- Additionally, SOSTDC1 enhances Wnt signaling and inhibits TGF-beta signaling, implicating it in the regulation of multiple developmental pathways.
- May play a role in endometrial receptivity for implantation and sensitization for the decidual cell reaction (by similarity).
- Subcellular localization: Secreted. It is highly expressed in the kidney and weakly in the lung, suggesting tissue-specific regulation.
- Post-translational modifications include glycosylation and disulfide bond formation, and multiple isoforms may arise via alternative splicing.

- For Western blot, the predicted molecular weight is ~23 kDa; glycosylation may shift the observed band. Use kidney lysates as a positive control given high expression.
