Biological Background: Sorbitol Dehydrogenase Function and Localization
- Sorbitol dehydrogenase (SORD) is a polyol dehydrogenase that catalyzes the reversible NAD-dependent oxidation of various sugar alcohols, with highest activity toward D-sorbitol, L-threitol, xylitol, and ribitol, converting them to D-fructose, L-erythrulose, D-xylulose, and D-ribulose, respectively. - SORD is a key enzyme in the polyol pathway, which interconverts glucose and fructose via sorbitol. This pathway is implicated in the pathogenesis of diabetic complications such as neuropathy and retinopathy under hyperglycemic conditions. - The enzyme also catalyzes the stereospecific oxidation of (2R,3R)-2,3-butanediol and, to a lesser extent, can oxidize L-arabinitol, galactitol, D-mannitol, and glycerol in vitro, but it cannot use NADP+ as a cofactor. Related references: PMID:3365415
- Subcellularly, SORD localizes to the mitochondrial membrane and is also found in cell projections such as cilia and flagella.
- Tissue expression is prominent in the liver, kidney, and epithelial cells of both benign and malignant prostate tissue, as well as in the epididymis at the protein level.
- Post-translational modifications include acetylation and phosphorylation, and the protein exists in alternatively spliced isoforms.
