Biological Background: Sorbitol Dehydrogenase Function and Localization

  • Sorbitol dehydrogenase (SORD) is a polyol dehydrogenase that catalyzes the reversible NAD+-dependent oxidation of sugar alcohols, with highest activity toward D-sorbitol, converting it to D-fructose. It cannot use NADP+ as electron acceptor. Related references: PMID:3365415
  • SORD is a key enzyme in the polyol pathway that interconverts glucose and fructose via sorbitol. This pathway is implicated in diabetic complications such as neuropathy and retinopathy induced by hyperglycemia. Related references: PMID:12962626, PMID:25105142, PMID:29966615
  • The enzyme may support sperm motility by using sorbitol as an alternative energy source. Related references: PMID:16278369
  • Substrate specificity includes D-sorbitol, L-threitol, xylitol, ribitol, and stereospecific oxidation of (2R,3R)-2,3-butanediol. It oxidizes primary alcohols poorly and does not act on ethanol.
  • Subcellular localization is primarily at the mitochondrial membrane and also in cell projections such as cilia and flagella. Tissue expression is highest in liver, kidney, prostate epithelium, and epididymis.
  • Structurally, it is a zinc-binding, NAD-dependent oxidoreductase that undergoes acetylation and phosphorylation. Alternative splicing produces multiple isoforms, and mutations in SORD are associated with hereditary neuropathy.
