Biological Background: Sonic Hedgehog (Shh) Function and Localization
- Sonic hedgehog protein (SHH) is a developmental morphogen that is synthesized as a precursor protein. The precursor undergoes autoproteolytic cleavage and cholesterol transfer in the endoplasmic reticulum to generate the N-terminal signaling domain (ShhN) and the C-terminal domain (ShhC). - The dually lipidated ShhN acts as a morphogen by binding to the patched receptor (PTCH1/PTCH2), which relieves inhibition of smoothened (SMO), thereby activating the SMO signaling pathway and transcription of target genes. Related references: PMID:10753901 PMID:24522195 PMID:24863049
- Shh is essential for a variety of patterning events during development, including central nervous system patterning (ventral cell fate induction), limb development (anterior-posterior axis), and axon guidance.
