Biological Background: Sonic Hedgehog (Shh) Function and Localization

  • Sonic hedgehog protein (SHH) is a developmental morphogen that is synthesized as a precursor protein. The precursor undergoes autoproteolytic cleavage and cholesterol transfer in the endoplasmic reticulum to generate the N-terminal signaling domain (ShhN) and the C-terminal domain (ShhC).   • Shh is essential for a variety of patterning events during development, including central nervous system patterning (ventral cell fate induction), limb development (anterior-posterior axis), and axon guidance.
