Biological Background: SOCS3 Function and Localization

  • SOCS3 is a member of the suppressor of cytokine signaling (SOCS) family, which forms a classical negative feedback loop regulating JAK/STAT cytokine signal transduction. Related references: PMID:16459330

  • It inhibits signal transduction by binding to phosphorylated tyrosine residues on transmembrane receptors such as gp130, LEPR, and G-CSF-R, and directly inhibits JAK2 kinase activity. Related references: PMID:16459330

  • SOCS3 functions as a probable substrate recognition component of an SCF-like ECS (Elongin BC-CUL2/5-SOCS-box) E3 ubiquitin-protein ligase complex, mediating ubiquitination and proteasomal degradation of target proteins. Related references: PMID:15601820

  • Negatively regulates cytokine signaling through the type II OSM receptor complex by interacting directly with JAK1 upon OSM stimulation. Related references: PMID:16459330

  • Widely expressed with high levels in heart, placenta, skeletal muscle, peripheral blood leukocytes, fetal and adult lung, and fetal liver and kidney; lower levels in thymus.

  • Contains an SH2 domain and a SOCS box domain, characteristic of the SOCS family, and is classified as a phosphoprotein involved in signal transduction inhibition.
