Biological Background: SOCS2 Function and Localization
- SOCS2 (Suppressor of cytokine signaling 2), also known as CIS2, SSI2, or STATI2, belongs to the SOCS family of cytokine-inducible SH2-containing proteins.
- SOCS2 functions as the substrate-recognition component of a cullin-5-RING E3 ubiquitin-protein ligase complex (ECS/CRL5). Through its SH2 domain, it binds phosphorylated substrates such as the growth hormone receptor (GHR) and erythropoietin receptor (EPOR), mediating their ubiquitination and subsequent proteasomal degradation, thereby acting as a critical negative regulator of growth hormone and cytokine signaling. - SOCS2 is predominantly localized in the cytoplasm.
- It is highly expressed in the heart, placenta, lung, kidney, and prostate, with particularly strong expression in pulmonary type II pneumocytes.
