Biological Background: SOCS1 Function and Localization
- SOCS1 (Suppressor of cytokine signaling 1), also known as JAB, SSI-1, or TIP-3, is a critical negative regulator of cytokine signaling pathways.
- It inhibits the JAK/STAT signaling pathway by binding to JAK kinases and cytokine receptors, thereby suppressing signaling of type I/II interferons, IL-2, IL-4, IL-6, and leukemia inhibitory factor (LIF).

- In immunohistochemistry (IHC-P), consider antigen retrieval methods (e.g., heat-induced epitope retrieval) and optimize primary antibody dilution based on tissue type. Nuclear and cytoplasmic staining patterns may be observed.
- For immunofluorescence/immunocytochemistry (IF/ICC), fix cells with paraformaldehyde and permeabilize with Triton X-100 to detect nuclear and vesicular SOCS1 localization; include isotype controls.
- As a polyclonal antibody, batch-to-batch variability may occur; perform pilot experiments and consider titrating the antibody for each new application or lot.
- Cross-reactivity with mouse and rat SOCS1 has been confirmed; however, always validate species-specific reactivity in your model system.
