Biological Background: SOAT1 Function and Localization
- Catalyzes the formation of fatty acid-cholesterol esters, which are less soluble in membranes than cholesterol, thereby regulating cholesterol homeostasis. Related references: PMID:16154994, PMID:16647063, PMID:32433613
- Plays a role in lipoprotein assembly and dietary cholesterol absorption. Related references: PMID:16154994, PMID:9020103
- Preferentially utilizes oleoyl-CoA ((9Z)-octadecenoyl-CoA) as a substrate, showing higher activity towards an acyl-CoA substrate with a double bond at the delta-9 position than towards saturated acyl-CoA or unsaturated acyl-CoA with a double bond at other positions. Related references: PMID:11294643, PMID:32433614
- Localizes to the endoplasmic reticulum membrane, the primary site of cellular cholesterol esterification.
- Post-translational modifications include acetylation and phosphorylation; the protein contains disulfide bonds and undergoes alternative splicing, yielding multiple isoforms.
- Functions in cholesterol storage and lipid droplet formation, and is a key enzyme in atherosclerotic foam cell development.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-130), which may detect both full-length and N-terminal isoforms; validate in relevant sample systems.
- For Western blot, expect a major band near 65 kDa; use lysates from human, mouse, or rat tissues/cells.
- In immunofluorescence, anticipate endoplasmic reticulum staining patterns consistent with its subcellular localization.
- For ELISA, consider pairing with a capture antibody or using the antibody itself as capture/detection after thorough optimization.
