Biological Background: SNX9 Function and Localization
- Sorting nexin-9, also known as SH3 and PX domain-containing protein 1 (SDP1) and SH3 and PX domain-containing protein 3A.
- Involved in endocytosis and intracellular vesicle trafficking, both during interphase and mitosis. Required for efficient progress through mitosis and cytokinesis, and normal cleavage furrow formation.
- Participates in clathrin-coated pit-mediated endocytosis and clathrin-independent, actin-dependent fluid-phase endocytosis. Also plays a role in macropinocytosis.
- Promotes internalization of TNFR and degradation of EGFR after EGF signaling.
- Stimulates the GTPase activity of DNM1, promotes DNM1 oligomerization, and activates the Arp2/3 complex via WASL, thereby reorganizing the F-actin cytoskeleton.
- Binds to membranes enriched in phosphatidylinositol 4,5-bisphosphate and promotes membrane tubulation; has lower affinity for phosphatidylinositol 3-phosphate-enriched membranes.
- Located at cytoplasmic vesicle membranes, cell membrane, clathrin-coated vesicles, trans-Golgi network, cell ruffles, and cytoplasm.
- Widely expressed, with highest levels in heart and placenta, and lowest in thymus and peripheral blood leukocytes.
