Biological Background: SNX3 Function and Localization
- SNX3, also known as Sorting nexin-3 or Protein SDP3, is a phosphoinositide-binding protein encoded by the SNX3 gene.
- It specifically binds phosphatidylinositol 3-phosphate (PtdIns(3)P) and can also interact with PtdIns(4)P, PtdIns(5)P, and PtdIns(3,5)P2, playing a key role in multivesicular body formation.
- In endosomal sorting, SNX3 functions as part of the SNX3-retromer complex to mediate retrograde transport of WLS from endosomes to the trans-Golgi network (TGN), distinct from the SNX-BAR retromer pathway. Related references: PMID:21725319, PMID:24344282, PMID:30213940
- It regulates phagocytosis in dendritic cells by controlling the recruitment of EEA1 to nascent phagosomes. Related references: PMID:23237080
- SNX3 is involved in iron homeostasis by regulating endocytic recycling of the transferrin receptor TFRC, directing the transferrin-TFRC complex to recycling endosomes.
- The protein promotes stability and cell surface expression of epithelial sodium channel (ENaC) subunits and contributes to neurite outgrowth in primary neurons.
- During Salmonella enterica infection, SNX3 promotes maturation of the Salmonella-containing vacuole and recruitment of LAMP1.
- Subcellular localization includes early endosomes, cytoplasmic vesicles, and phagosomes; post-translational modifications include acetylation, methylation, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal 162 amino acids, including the PX domain responsible for phosphoinositide binding; this region is well conserved across human, mouse, and rat.
- For Western blotting, the predicted molecular weight is ~19 kDa; consider that post-translational modifications or isoforms may cause slight shifts. Validate in relevant cell or tissue lysates.
- The polyclonal nature may provide sensitive detection in ELISA; for sandwich ELISA, pairing with a monoclonal capture antibody targeting a non-overlapping epitope is recommended.
- Cross-reactivity with mouse and rat has been confirmed by the supplier, making it suitable for rodent model studies. Always include appropriate controls to verify specificity in your system.
